GeneFunction Connors et al. (1999) generated a tolloid transgene under the control of the inducible hsp70 promoter and demonstrated the role of tolloid (mfn) in completion of gastrulation. Chimeric studies indicate the requirement of tld for ventral (posterior)tail bud expression. The dorsal (anterior) tail bud expression domain of tld lies in closer proximity to chd expressing cells in the tail bud. Using the heatshock-inducible tld transgene, they revealed the function of chd in tail patterning both during and after gastrulation. Thus, fine modulation of BMP signaling levels through the negative and positive actions of Chordin and Tolloid, respectively, patterns tail tissues.Herpin et al. (2007) cloned a gene encoding a tolloid homologue from the bivalve mollusk Crassostrea gigas. They determined the intron-exons structure of the gene encoding C.gigas tolloid and have compared it with those of homologous genes from both protostomes and deuterostomes. They analyzed the functionality of oyster tolloid in zebrafish embryo and showed the ventralization of zebrafish embryos at 24 h post fertilization, when the protein is overexpressed.

DisordersAndMutations Blader et al. (1997) discovered the role of tld in cleaving chd invitro but not noggin or follistatin. It can also inhibit chd function in vivo through the mechanism by which tld increases BMP activity.Connors et al. (1999) injected tolloid mRNA into six dorsalized mutants out of which they rescued only one mutant, mini fin that was then demonstrated to encode tolloid gene. Minifin/tolloid activity is required for patterning ventral tissues of the tail: the ventral fin, the ventroposterior somites and vasculature, and is expressed throughout gastrulation in a ventral-animal domain, as well as dorsally and ventrally around the margin. In post-gastrula stages, it is expressed dorsally and ventrally in the tail bud with its dorsal limit of expression directly abutting chd expression.Jasuja et al. (2006) identified and mapped the zebrafish BMP1 gene. Knockdown analysis of BMP1 resulted in a mild tail phenotype, however, simultaneous knockdown of mini fin and bmp1 resulted in severe dorsalizaion resembling the swirl (swr) and snailhouse (snh) phenotypes. Thus they concluded bmp1 and mfn gene products that functionally overlap and are together responsible for a key portion of the chordin processing activity necessary to form zebrafish dorsoventral axis.
